A strong toxin known as tetrodotoxin (TTX) binds exclusively to voltage-gated sodium channels. Action potential (AP) generation and propagation are prevented by TTX binding because it physically restricts the flow of sodium ions through the channel. Varied sodium channel isoforms have different binding affinities for TTX.

These variations are brought about by substitutions of amino acids. Different species are given TTX resistance thanks to these substitutions. However, tetrodotoxin resistance may have a negative impact on performance due to alterations in the biophysical characteristics and/or ion selectivity of the sodium channels that are TTX resistant.
